Human hemoglobin is used as a marker for human blood. Although tandem mass spectrometry is the dominant methodology used in proteomics research, peptide mass fingerprinting, given its instant applicability, may be useful for screening animal species, as the amino acid sequences of hemoglobin from various animals differ. In this study, hemoglobin containing solutions were easily purified from humans, Japanese macaques, bears, cattle, goats, sheep, sika deer, pigs, wild boars, dogs, cats, and nutrias were digested by trypsin, and subjected to database search using Mascot. The article states “We have developed the application of commercially available reagents for simply purifying hemoglobin from a wide range of animal species. …Blood, saliva, urine, semen, and sweat (20 μL); filter paper from which sweat was collected; and 0.5, 1, 2, and 5 μL of pooled blood samples were treated with HemogloBind™ (BIOTECH SUPPORT GROUP, NJ, USA) to obtain hemoglobin solutions following manufacturer’s protocol. …Hemoglobin was then dissociated according to the protocol to obtain a purified solution. These eluates were exchanged with water and concentrated to a final volume of 30−50 μL…To remove contaminating proteins from the solution, 5 mg of HemoVoid™ (BIOTECH SUPPORT GROUP) was used on a 1/10 scale of the protocol.“. The SDS-PAGE of purified solutions is from different animal bloods using a combination of the two hemoglobin reagents; one that binds - HemogloBind, and one that voids hemoglobin out, HemoVoid. In conclusion, this method may be useful for estimating animal species and identifying blood in forensic science.


I am pleased to see that this is the second reported use of HemogloBind™ for purifying Hemoglobin, even though that was not initially its intended purpose. The previous report is documented in one of our case studies that is available on our website. These two reports demonstrate that HemogloBind™ by itself or in combination with HemoVoid™ can provide sufficient purity for mass spectrometry analysis of Hemoglobin either for forensic or point mutation analysis.
