Cleanascite™ for removing serum lipids from anti-H5 antibody MONMOUTH JUNCTION, N.J. — In a published article in the Journal of Biosensors and Bioelectronics titled, “A nanobeads amplified QCM immunosensor for the detection of avian influenza virus H5N1” scientists successfully used Cleanascite™ for removing serum lipids from rabbit anti-H5 after ammonium sulfate precipitation. The sample monoclonal antibody is produced by intraperitoneal immunization of rabbits with concentrated purified virus preparation containing hemagglutinin (HA) protein of influenza A virus. This method is described by Kohler and Milstein. Often the rabbit anti-H5 is clarified from mouse ascitic fluid and Cleanascite™ from Biotech Support Group is an ideal reagent for delipidation treatments allowing for downstream processing of large-scale therapeutic proteins, enzymes and monclonal antibodies. Moreover, Cleanascite™ does not bind to DNA, RNA, enzymes and proteins. The samples were centrifuged and the immunoglobulins in the supernatant were precipitated by gradual addition of ammonium sulfate to 45% saturation. The IgG fraction was collected by centrifugation, dissolved in PBS, and dialyzed. The antibody concentration at 280nm was calculated by absorbance. Through this article, polyclonal antibodies against AI H5N1 virus surface antigen HA (Hemagglutinin) were immobilized on the gold surface of the QCM crystal through self-assembled monolayer (SAM) of 16-mercaptohexadecanoic acid (MHDA). This monoclonal antibody can be used for hemagglutination inhibition (HI) assays to provide antigenic characterization of the influenza A viruses of the H5 HA subtype. The major functions of HA include receptor-binding and fusion activities,. The rabbit anti-H5 monoclonal antibody works for sample preparation in virus neutralization assays (in cell culture and in embryonated chicken eggs), ELISA, immunoprecipitation, immunohisto-chemistry and western blotting. Scientists successfully used magnetic nanobeads based QCM immunosensor technology to detect AI H5N1 virus in tracheal swab samples and to distinguished h AI H5N1 from AI subtypes H3N2, H2N2, and H4N8.
