Nucleated red blood cells (RBCs) of fish have, in the last decade, been implicated in several immune-related functions, such as antiviral response, phagocytosis or cytokine-mediated signaling. Label-free shotgun proteomic analyses were carried out for in silico functional pathway profiling of rainbow trout RBCs. The article states “The cytosolic fraction, approximately 300 μL, was depleted of hemoglobin using HemoVoidTM kit (Biotech Support Group, Monmouth Junction, NJ, USA), in accordance with the manufacturer’s instructions”. The authors conclude that by combining transcriptome- and proteome- sequencing data in silico, novel and striking immune functional networks for rainbow trout nucleated RBCs, which are mainly linked to innate and adaptive immunity. These preliminary findings further implicate nucleated RBCs in immune function, such as antigen presentation and leukocyte activation. “This article illustrates that our proteomic enrichment products work very efficiently for all species, even non-mammals, as they do not rely on antibodies. The ability to enrich for the underlying sub-proteome from the cytosolic fraction of red cells is a unique feature of HemoVoid™.
