New research on Albusorb™ for depletion of albumin from mouse, rat serum and plasma samples MONMOUTH JUNCTION, N.J. — Serum albumin can constitute more than 55% of total serum protein, obscuring other proteins and making the resolution of lower abundant proteins difficult. Biotech Support Group offers two new products for albumin depletion: AlbuSorb™ and AlbuVoid™.AlbuSorb™ binds albumin allowing serum proteins to flow through; while AlbuVoid™ allows the albumin to flow through, the serum proteins bind and elute. Removing albumin enhances the detection of low abundance proteins.AlbuSorb™ and AlbuVoid™ come from a class of novel surface-coupled elastomeric polymers. By combining elements of polymer composition, surface architecture and charge properties, reactivity is governed along a flexible polymer chain. AlbuSorb™ and AlbuVoid™ are economically advantageous in high throughput operations.

Leptin expression is detectable by Western blot analysis only when the total protein concentration of the serum sample is >0.2 mg/ml. “After depleting albumin from serum by AlbuSorb (Biotech Support Group, LLC, North Brunswick, NJ) according to the manufacturer’s protocol, the serum samples were diluted 2-fold and denatured in the presence of sodium dodecyl sulfate and 2-mercaptoethanol by heating at 100°C for 5 min.” A total of 13.9 μg proteins in each sample were electrophoretically transferred to 2 nitrocellulose membranes (Amersham Bioscences Piscataway, NJ). Nonspecific binding on the membranes was minimized with 5% skim milk at room temperature for 1 h, followed by incubation overnight at 4°C in 5% skim milk containing the polyclonal rabbit antileptin antibody (1:200 dilution, A-20, Santa Cruz Biotechnology, Inc., Santa Cruz, CA).
