A comprehensive analysis of the humoral immune response (the immunome) has potential to greatly impact research across numerous fields. For example, serum autoantibodies against tumor-associated antigens have recently emerged as early stage biomarkers for different types of cancers. Most autoantibody profiling work has been based on the reactivity of unbound antibodies towards antigens produced by a variety of strategies (i.e., cDNA libraries, phage display).

An alternative approach is based on the identification of Ig-bound antigens using Liquid Chromatography coupled to Mass Spectrometry (LC-MS). Such determination of antigens complexed with antibodies at a proteome scale is critical to understanding adaptive responses in the context of infection, autoimmunity, and cancer.

Human serum immunoglobulins comprise several classes IgG, IgA, IgM, IgD & IgE. IgG is the predominant human immunoglobulin class in plasma and comprises four subclasses; ~60% are IgG1, followed by ~30% IgG2, ~7% IgG3 and ~3% IgG4. To date, most of the circulating antibody complex research has been focused on IgG as the efficiency of recovering a representative pool of IgG antibodies is well established. Generally for human serum/plasma, Protein A binds with high affinity to IgG1, IgG2, and IgG4, but poorly to IgG3. Among the four IgG subtypes in mice, Protein A has the weakest affinity for IgG1 while Protein G has affinity for all four IgG subclasses. Neither Protein A or G bind particularly well towards IgA, IgM, IgD or IgE.

Nevertheless, the ability to enrich circulating immune complexes from sera or plasma from both animals and humans with high yield and without selective loss of isotypes or subclasses can provide more comprehensive profiles. NRicher™ Ig can provide such enrichment for all immunome profiling methods. For antigen reactivity profiling, elution conditions are mild (pH 9-10), and preserve functionality. For antigen identification, bound proteins can be digested on-bead, with seamless integration to LC-MS analysis.

Thearticle states we used affinity beads (NuGel PROspector)to pre-enrich Toxoplasma gondii lysate to capture five distinctsubproteomes. [Note: NuGel PROspector beads are now part of the NRicher™ platform.]. When comparing the 5 differentsubproteomes, there is clearly different selection biases amongst the5 surface chemistries. Also, many of the proteins observed from the NRicher™ beads, were not observed in the Ion exchangefractions demonstrating the importance of combining different modes(ionic, hydrophobic, etc.) of separation to alter selectionproperties, and consequently improving overall proteome coverage.

Theneed for new biomarkers to support personalized healthcare, hasfostered numerous proteomic innovations. Still, a number ofchallenges remain. Do BSG's products work for all species?

Yes, as our products are not derived from antibodies, they are species agnostic, and have applied to humans, mice, rats, guinea pigs, goats, cows, pigs, even frogs and fish.

Can you supply bulk material only (from the kit-based products) for large volume orders?

Yes, we can adjust all of our kit products, to supply only the beads. For large orders, whether kit or bulk material, we provide special discounted quotations. Please inquire.

Can the protocol be scaled to accommodate different amounts of either volume or mass?

Yes, we provide general guidelines for use, but the protocols can usually be adjusted to accommodate either larger or small starting samples, or larger or smaller starting concentrations. We recommend first contacting us for guidance in adjusting the protocol.

Can the products be adapted to high throughput formats and automation?

Although we do not have an off-the-shelf product for 96-well formats currently, we have ongoing collaborations that can provide such formats. Please inquire.

Are the products compatible with detergents?

This has not been fully investigated, but we do not recommend use of SDS. Low concentrations, 0.1 – 0.5% of non-ionic detergents have been used with some of our products, but to what extent they alter separation performance, is unknown. If non-ionic detergents are to be used, generally we recommend that they be of no greater than 0.1% before applying to the beads. So for tissue lysates for example at higher %, dilute the sample before application.

What is the stability and shelf life of the product?

The products have a shelf life typically 6 months to 1 year. We ship at room temperature, but recommend storage at 4°C upon receipt. Do not freeze.

Do the products contain any biologically derived material?

All of our products are derived chemically, not biologically, with one exception. The "PLUS" products contain recombinant Protein A.
