Cleanascite™ Cited in Nature Study of Neuronal Proteostasis with Age
For proteomics of homogenized brain tissue, the article states “Lipids were removed by the addition of 10 µl Cleanascite per 40 µl homogenate and incubation with constant agitation on a thermomixer set to 1,500–2,000 rpm for 10 min.”. Upon further bead-based pulled down, samples were analyzed by Liquid-Chromatrography – Mass Spectrometry (LC-MS/MS) for proteomic comparisons and TMT-labeled quantifications.
In part through the use of Cleanascite™, combining protein aggregate isolation techniques with neuronal BONCAT labelling helped define the neuronal aggregome, a catalogue of neuronal proteins that contribute to protein aggregates in the aged brain. LC–MS analyses enabled identification of 1,726 neuronal proteins present in aggregates in the aged brain, 392 of which have been identified in aggregates of human brains from old individuals. These findings reveal the substantial loss of neuronal proteome maintenance with ageing, which could be causal for age-related synapse loss and cognitive decline.
“Here
is another excellent example of how Cleanascite™ can be used to
clear lipids from a lipid rich tissue such as brain, and that its
excellent selectivity for removing lipids makes it ideally suited for
further proteomic analysis by LC-MS/MS.” stated Dr. Swapan Roy,
President and Founder of Biotech Support Group. For
