— Biotech Support Group (BSG) reports on itsproprietary lipid removal reagent, Cleanascite™, in a mousemodel study of neuronal protein maintenance and the accumulation ofprotein aggregates with ageing.


Cleanascite™ Cited in Nature Study of Neuronal Proteostasis with AgeIn a Nature article led by Tony Wyss-Coray at StanfordUniversity, entitled Ageingpromotes microglial accumulation of slow-degrading synapticproteins”,researchers leveraged robust in vivo models to examine proteindynamics in a cell-specific manner in mammals. These models were usedto study key features of neuronal proteostasis dynamics with ageingto provide detailed insights into the decline of neuronalproteostasis with age.


Forproteomics of homogenized brain tissue, the article states “Lipidswere removed by the addition of 10 µl Cleanascite per 40 µlhomogenate and incubation with constant agitation on a thermomixerset to 1,500–2,000 rpm for 10 min.”. Upon further bead-basedpulled down, samples were analyzed by Liquid-Chromatrography – MassSpectrometry (LC-MS/MS) for proteomic comparisons and TMT-labeledquantifications.


Inpart through the use of Cleanascite™, combining protein aggregateisolation techniques with neuronal BONCAT labelling helped define theneuronal aggregome, a catalogue of neuronal proteins that contributeto protein aggregates in the aged brain. LC–MS analyses enabledidentification of 1,726 neuronal proteins present in aggregates inthe aged brain, 392 of which have been identified in aggregates ofhuman brains from old individuals. These findings reveal thesubstantial loss of neuronal proteome maintenance with ageing, whichcould be causal for age-related synapse loss and cognitive decline.
