FraserDD, Roy S, Kuruc M, Quintero M, Van Nynatten LR, Cepinskas G, ZhengH, Soherwardy A and Roy D (2025) Functionalmass spectrometry indicates anti-protease and complement activityincrease with COVID-19 severity. Exp.Biol. Med. 250:10308.doi: 10.3389/ebm.2025.10308. Investigations on some innate immunityproteins can yield misleading information, as investigators mostoften rely on static measurements and assume a direct correlation tofunction. However, function is often not directly proportional toprotein abundance, and mechanistic pathways are interconnected andunder constant regulatory control. In this study, we used functionalmass spectrometry to measure anti-protease and complement activity inplasma obtained from coronavirus disease 2019 (COVID-19) patients. The target peptides were selected to report specific regions of theprotein(s) that infer functionality. Taken together, tryptic peptidesselected from functional regions thus can serve as surrogates forreporting functional activity. For Albumin & IgG depletion, thearticle states “The clarified serum was loaded onto the conditionedAlbuSorb™PLUS beads…”, prior to LC-MS/MS analysis.
Grantz,Jillian M., et al. "Theplatelet and plasma proteome and targeted lipidome in postpartumdairy cows with elevated systemic inflammation." ScientificReports 14.1(2024): 31240. Unregulated, systemic inflammation negatively impactshealth and production in dairy cows. Soluble mediators and plateletshave been studied for their expansive role in mediating inflammation. The objectives of this study were to compare the plasma oxylipin andendocannabinoid profiles, and the platelet and plasma proteomicprofiles of healthy cows to cows experiencing elevated systemicinflammation as indicated by plasma haptoglobin (Hp) concentrations. The article states “Plasma samples weredepleted of albumin using AlbuSorb™ Plus Albumin and IgG depletionKit(APK285-20, BioTech Support Group, Monmouth Junction, NJ), followingthe manufacturer’s instructions.” In total, 1,016 proteins wereidentified in the plasma proteome. Proteomic outputs from plasmarevealed 24 proteins to be different between high andlow-inflammation groups, including proteins involved in autophagy andimmune mediation.
Jordan,Christine KI, et al. "SymbioticFirmicutes establish mutualism with the host via innate tolerance andresistance to control systemic immunity." CellHost & Microbe (2023). The intestinal microbiota regulates immunity across organ systems.Which symbionts control systemic immunity, the mechanisms they use,and how they avoid widespread inflammatory damage are unclear.Despite systemic penetration of Firmicutes, immune homeostasis ismaintained through feedback control whereby macrophages clearpolymeric glycoconjugates from peripheral tissues. Smallerglycoconjugates evading this clearance mechanism are toleratedthrough sequestration by albumin, which acts as an inflammatorybuffer constraining their immunological impact. To determine this, the article states, “AlbuSorb(Biotech Support Group) was used to deplete albumin from mouse plasmaas per manufacturer instructions.” “We found that complete humanand murine plasma activated our NOD2, TLR2, and TLR4 reporter cellsandthat post-albumin depletion, plasma from both species activatedreporter cells less potently, indicating that albumin sequesters circulating cell wallglycoconjugates.”

Increased levels of apolipoprotein CIII (apoCIII),result in obesity-related metabolic derangements. Using mice, theresearchers investigated mechanistically whether lowering orpreventing high-fat diet (HFD)– induced increase in apoCIII,protects against the detrimental metabolic consequences. For Westernblotting determination of circulating apoCIII, the article states,“plasmawas albumin depleted using AlbuSorbaccording to the manufacturer’s protocol (Biotech Support GroupLLC) and resuspended in 0.1% (v/v) trifluoroacetic acid.”

Matsuzawa-Ishimoto,Yu, et al. "Theγδ IEL effector API5 masks genetic susceptibility to Panethcell death." Nature (2022):1-8. Lossof Paneth cells and their antimicrobial granules compromises theintestinal epithelial barrier and is associated with Crohn’sdisease. Non-classical lymphoid cells, broadly referred to asintraepithelial lymphocytes (IELs), intercalate the intestinalepithelium. The identification of mediators that coordinate crosstalkbetween specific IEL and epithelial subsets could provide insightinto intestinal barrier mechanisms in health and disease. Here weshow that the subset of IELs that express γ and δ T cell receptorsubunits (γδ IELs) promotes the viability of Paneth cells deficientin the Crohn’s disease susceptibility gene ATG16L1.Forimmunoblotting of intestinal organoid supernatants, the articlestates “…albumin was depleted with AlbuSorb…according to the manufacturer’s instructions”.
