Cleanascite™ Cited in Study of Chemical Interference of Thyroid Hormone MetabolismInthe article led by Jermaine Ford at the US Environmental ProtectionAgency, entitled “ToxicologicalProfile of Chemical Interference of Thyroid Hormone Metabolism ViaDeiodinase Enzymes In a Rodent Model”,researchers leveraged rat models exposed to iopanoic acid (IOP), apotent in vitroinhibitor of Deiodinases (Dio), a family of metabolizing enzymescontrolling systemic and local availability of the major thyroidhormones (TH). To measure these hormones, the investigators optimizeda liquid chromatography-tandem mass spectrometry (LC-MS/MS) method tocorrelate the effects of IOP on Dio activity in liver to TH profilesin serum. The article states “Liver samples were homogenized… .Internal standard containing 13C labeled 3,3’-T2, T3, rT3, and T4at 100 ng/mL along with 50 μL of Cleanascite™. Upon furthersolid-phase extraction, samples were analyzed byliquid-Chromatrography – Mass Spectrometry (LC-MS) for THmetabolite comparisons.

Quantitativeanalysis using a selective and sensitive LC-MS/MS-based assay fordeiodinase inhibition in the liver of exposed animals revealed robustsuppression of Dio1 and Dio3 activity in hepatic microsomes in apattern consistent with serum profiles. As such, this study providesa comprehensive characterization of the in vivo effects ofchemicals that interfere with the metabolism of TH by hepaticdeiodinases, enzymes that largely control the serum concentrations ofTH. In part through the addition of Cleanascite™, optimization ofthe LC-MS/MS based assay described here permitted precise estimatesof deiodinase action relative to alternative approaches and offersnew insights into the broader implications of chemical interference.
