Cleanascite™ Cited in Study of Chemical Interference of Thyroid Hormone Metabolism

Cleanascite™ Cited in Study of Chemical Interference of Thyroid Hormone MetabolismIn the article led by Jermaine Ford at the US Environmental Protection Agency, entitled “ Toxicological Profile of Chemical Interference of Thyroid Hormone Metabolism Via Deiodinase Enzymes In a Rodent Model ”, researchers leveraged rat models exposed to iopanoic acid (IOP), a potent in vitro inhibitor of Deiodinases (Dio), a family of metabolizing enzymes controlling systemic and local availability of the major thyroid hormones (TH). To measure these hormones, the investigators optimized a liquid chromatography-tandem mass spectrometry (LC-MS/MS) method to correlate the effects of IOP on Dio activity in liver to TH profiles in serum. The article states “Liver samples were homogenized… . Internal standard containing 13C labeled 3,3’-T2, T3, rT3, and T4 at 100 ng/mL along with 50 μL of Cleanascite™. Upon further solid-phase extraction, samples were analyzed by liquid-Chromatrography – Mass Spectrometry (LC-MS) for TH metabolite comparisons.

Quantitative analysis using a selective and sensitive LC-MS/MS-based assay for deiodinase inhibition in the liver of exposed animals revealed robust suppression of Dio1 and Dio3 activity in hepatic microsomes in a pattern consistent with serum profiles. As such, this study provides a comprehensive characterization of the in vivo effects of chemicals that interfere with the metabolism of TH by hepatic deiodinases, enzymes that largely control the serum concentrations of TH. In part through the addition of Cleanascite™, optimization of the LC-MS/MS based assay described here permitted precise estimates of deiodinase action relative to alternative approaches and offers new insights into the broader implications of chemical interference.
