Inan example, pancreatic islets were isolated from male 8-14 week oldC57B16/J mice (Jackson Laboratory, ME) and cultured in various mediaconditions for 3 days.
Thepatent states “Forlipid removal, serum was treated with Cleanascite reagent (BiotechSupport Group, Monmouth Junction, N.J.) prior to islet culture at a1:1 ratio according to the vendor's protocol.”.The example supports that increasedamino acids, but not lipids and other soluble factors, selectivelyincreased rapamycin-sensitive α-cell proliferation.
